Protein Complex Characterization Group

The Cancer Research Technology Program (formerly the Advanced Technology Program) at NCI-Frederick has extensive expertise in protein expression, isolation, and characterization.  For example, the Protein Characterization Laboratory is fully equipped with state-of-the-art mass spectrometry and other protein detection instrumentation.  To facilitate the identification and characterization of protein complexes, the CRTP has assembled a group of scientists that is dedicated to assisting CCR investigators with the design and execution of expression and purification strategies to define the composition of protein complexes in mammalian cells.

Established Technologies:

  • Preparation of cell lines transfected with expression clone
  • Optimization of immunoprecipitation conditions for complex isolation
  • MS characterization of isolated complex metabolites from human biofluids and tissue
  • Validation of protein interactions

Protein Mass Spectrometry Group

The PCL Mass Spectrometry Center is equipped with the latest in mass spectrometry (MS) instrumentation for proteomic and metabolomic analysis. Areas of expertise include protein complex characterization, global and target protein quantitation, and identification of post-translational modifications. The Center also provides educational and training opportunities for scientists on the use, interpretation, and applications of MS technologies.

Protein Chemistry and Molecular Interactions Group

The group develops and maintains unparalleled abilities in surface plasmon resonance (SPR) spectroscopy and fluorescence and has complementary protein chemistry techniques for protein identification, including mass spectrometry and classic Edman protein degradation sequencing. The group has state-of-the-art MALDI-TOF mass spectrometry with image and high mass detection capabilities and provides HPLC purification and quality control of protein and oligonucleotides.
