10X Genomics Chromium Single Cell Sequencing

Droplet-based single cell sequencing enables a relatively robust and accessible platform to examine multiple molecular modalities at the resolution of the single cell. On the 10x Genomics Chromium platform, hundreds to thousands of cells or nuclei suspensions can be profiled in parallel. In addition to or in combination with standard single cell or single nuclei RNA sequencing, modalities that can be profiled include full-length T and B cell immune repertoire profiling, cell surface protein labeling, expressed lineage barcodes, and CRISPR functional genomic screens. A new probe-based version of the Chromium transcriptional profiling assay allows the profiling of fixed cells, including those isolated from FFPE blocks.

10X Genomics Visium Spatial Transcriptomics

Visium FFPE v2 is sequencing-based spatial profiling technology developed by 10x Genomics. This assay can take mouse or human tissue sections on normal glass slides as input and will provide a whole transcriptome survey of the gene expression profiles across the tissue section. Resulting data will be gene expression counts value for each spatial spot (55 micron diameter, 100 micron from each other), which is overlayed over the original tissue image. While the current version does not provide single cell resolution, the whole transcriptome analysis across the entire interrogated tissue region will provide a relatively unbiased survey of the spatially organized gene expression in a given sample. Upcoming versions will improve upon resolutions.

10X Genomics Xenium In Situ Sequencing

Xenium is a high-resolution, imaging-based in situ spatial profiling technology from 10x Genomics that allows for simultaneous expression analysis of RNA targets (currently in range of 100’s) within the same tissue section. This assay can identify the location of target transcripts within the tissue, providing a single cell resolution map of expression patterns of all genes that are included in the selected probe panel. Researchers can choose between off-the-shelf panels of genes designed to identify cells types and interrogate specific phenotypes, add on 50-gene or 100-gene panels to these off-the-shelf panels, or design a full custom standalone panel to specifically tailor for their tissue and research questions.
